Amyloid Plaque Fighting Synergy

Recent studies have demonstrated that resveratrol reduces the incidence of age-related macular degeneration, Alzheimer’s disease (AD), and stroke, while melatonin supplementation reduces the progression of the cognitive impairment in AD patients. In a new investigation, the co-administration of melatonin and resveratrol was studied to determine whether there were any synergistic effects on their neuroprotective properties against amyloid-β (Aβ)-induced neuronal death.

Co-treatment on Aβ1-42-induced* cell death was measured an assay along with other tests to determine the intracellular signaling mechanism involved in these synergic effects.


* Aβ 42 is the Aβ isoform most associated with diseased states.


While co-treatment did not exert significant synergistic effects on the recovery of the Aβ1-42-induced total glutathione levels depletion or on the inhibition of Aβ1-42-induced glycogen synthase kinase 3β activation, Aβ1-42 treatment increased AMP-activated protein kinase activity, which is associated with subsequent neuronal death. Together, melatonin and resveratrol treatment inhibited the phosphorylation of AMPK, an enzyme that plays a role in cellular energy homeostasis.
